Rapid Technique for Flexirubin-type
pigments test:
Ø Bacteria consist of several types of
pigments. Flexirubin type pigments is one of the important chemosystematic
markers used for species level differentiation. This type of pigment is
identified in Flexibacter and Cytophaga strains and in Flavobacteria which have
low G+C mol% value.
Ø For Screening the presence of
flexirubin type pigments in bacteria, following steps can be performed:
1.
Flood
the culture plate with 20% KOH solution
2.
Note
the rapid color shift from yellow or orange to red, purple or brown color.
3.
Decant
the excess KOH and then add acetic acid to observe the revert of initial color.
Ø Note that when color changes after
adding 20% KOH and again revert back to original color after adding aceti acid
indicates presence of flexirubin type pigments. When color do not changes after
adding 20% KOH as well as do not revert back to original color after adding
acetic acid indicates absence of Flexirubin type pigments in the bacteria.
